The shrimp farming industry is constantly under threat due to the outbreak of infectious diseases and environmental problems. The infections caused by Vibrio species is referred as vibriosis and could cause serious infections thus decreased the amount of production. Microbiological assessment of aquaculture systems useful to determine the potential success or biological contamination in aquaculture industry. The aim of the research is to provide monitoring and risk mitigation assessment from three sampling locations in Sarawak, Malaysia; Location 1 (Telaga Air), Location 2 (Santubong), and Location 3 (Tanjung Manis), in regard with the distribution of Vibrio species. A total of thirty-two water samples and thirty-two sediment samples at two shrimp ponds (Pond A and Pond B) from three sampling locations were collected in this study from a complete cycle of stocking to harvest process which from the 1st first day of Culture (DOC) until 98th DOC. Physio-chemical analysis of the shrimp ponds were recorded during the sampling. The average pH of pond water for most farms was range between 6.5 to 8, which is in optimum range for the shrimp growth range. The temperature of pond water recorded during sampling for all three locations was range between 28 ℃ to 35 ℃. The salinity of the pond water for most farms was range between 15 to 27 ppt. Three Vibrio species were identified using multiplex PCR with primers used are Vibrio parahaemolyticus were amplified at 5’ AGCTT ATTGG CGGTT TCTGT CGG and 5’ CKCAA GACCA AGAAA AGCCG, Vibrio alginolyticus were amplified at 5’ ACGGC ATTGG AAATT GCGAC TG and 5’ TACCC GTCTC ACGAG CCCAA G and Vibrio cholera at 5’ CAAGC TCCGC ATGTC CAGAA GC and 5’ GGGGC GTGAC GCGAA TGATT. The product size for 297 bp. 199 bp and 154 bp receptively. The study was carried out by analyzing fourteen commonly used antibiotics in aquacultures industries by using Antimicrobial Susceptibility Test (AST). The iv results show that Location 1 (Telaga Air) have the lowest log cfu/ml with the average of 3.85 log cfu/ml, followed by Location 2 (Santubong); 3.91 log cfu/ml, and Location 3 (Tanjung Manis); 4.02 log cfu/ml. However, for distribution of Vibrio species in sediment samples showed the Location 1 (Telaga Air) and Location 2 (Santubong) have the same average 3.95 log cfu/ml, and Location 3 (Tanjung Manis) with 3.97 log cfu/ml. The amplification of Vibrio parahaemolyticus was high in all sampling locations, while, for Vibrio alginolyticus at Location 1 (Telaga Air), and for Vibrio cholerae at Location 2 (Santubong). Vibrio parahaemolyticus were highly amplified from both water and sediment samples, while Vibrio alginolyticus and Vibrio cholerae were highly amplified from sediment samples only. A total of sixty of positive isolates were tested with antibiotic susceptibility test (AST) and the results showed that penicillin recorded highest resistance percentage (40%) towards tested isolates in all three sampling locations, followed by streptomycin (30%). The MAR index in this study ranged from 0.35 to 0.65. This study has contributed useful monitoring data on the distribution of Vibrio species in aquaculture shrimp farms in Sarawak. Furthermore, this study has suggested effective shrimp pond management strategies to help in controlling the numbers of Vibrio species in aquaculture shrimp farms. Daily monitoring of physio-chemical and microbiological characteristics of shrimp pond are essential to keep tract for troubleshooting problems and easy reference. Monitoring was recommended for at least twice daily (morning and evening) to maintain the optimum conditions for growth and to avoid any drastic changes of the parameters.
